The southern terminus of the route is at an intersection with NY 36 in the hamlet of Mumford within the town of Wheatland. Its northern terminus is at a junction with NY 31 in the city of Rochester. The route follows the Genesee River and its tributaries for its entire length and passes through the village of Scottsville.
